The human embryonic stem cell-derived retinal pigment epithelial cells (hESC-RPE) hold great potential as the viable cell source for treating degenerative retinal diseases. However, further improvements are required to enhance the transplantation efficiency and viability of hESC-RPE. This study aimed to explore a novel form of carrier called Gelatin-methacryloyl / Hyaluronic acid-methacryloyl microspheres coated with fibrin (Fib@GHMS), which may efficiently load and distribute hESC-RPE while also promoting visual function recovery after subretinal injection. We generated injectable Fib@GHMS with consistent particle size via microfluidics. Subsequently, we evaluated its morphology, chemical composition, and cytotoxicity by scanning electron microscopy, Fourier transform infrared spectroscopy, and biocompatibility assessments. The findings demonstrated that Fib@GHMS exhibited desirable biocompatibility and displayed enhanced adhesion and proliferation of hESC-RPE. Our work also provided a theoretical and experimental basis for future advancements in this field.
