Diisononyl phthalate (DINP) is a high-molecular-weight phthalate, and has been recently introduced as DEHP substitute and commonly used in a large variety of plastic items. The fat tissue is an important target for DINP exposure, however, very little is understood about its toxicity and mechanism(s) in adipocyte cells. Therefore, the present work aimed to investigate the role of DINP in adipogenesis using 3T3-L1 preadipocytes. The 3T3-L1 preadipocytes were treated with 50 micromolar of DINP, and the global gene expression was analyzed based on the sequencing platform, BDISEQ-500 with a read length of 50 bp. The RNA-Seq results revealed that the PPARγ pathway and lipid metabolism are the most significantly affected function for under DINP exposure.
