The main objective of the sudy was to characterize the functions of Drosophila Hepatocyte Nuclear Factor 4 (dHNF4) in midgut enterocytes. For this, we expressed a UAS-dHNF4 RNAi transgene under the control of mex-GAL4, which is active specifically in midgut enterocytes. We then collected adult male midguts, extracted total RNA and performed mRNA-seq. Our data demonstrate that dHNF4 acts as a master regulator of intestinal lipid metabolism in Drosophila. Total RNA was extracted from 20 midguts of adult male flies ( controls - genotype: w1118/Y ; ; UAS-dHNF4 RNAi NIG9310-R2/+), and of adut male flies expressing a UAS-dHNF4 RNAi transgene under the control of the enterocyte-specific mex-GAL4 driver (genotype: w/Y ; mex-GAL4/+; UAS-dHNF4 RNAi NIG9310-R2/+). mRNA sequencing was performed on 5 biological replicates per genotype. Adult male flies were fed a yeast only diet and dissected 7 days after eclosion.
