Study Purpose: To investigate the changes of lipid metabolites in Drosophila fat body due to premature or inhibited autophagySample Details: The larvae were collected for more than 20 mg in each sample from WT1118, DmAtg1Δ3D, and ppl-GAL4>UAS-DmAtg1:DmAtg13 at wandering (W), white prepupa (WPP), and 3 h after white prepupa (3WPP) stages. Each set of treatments contained six biological replicates.Instrumentation and Data Processing: The Drosophila larvae in each sample were homogenized and extracted for lipids by 1 mL extract solution containing methyl tert-butyl ether and methanol in the volume ratio of 3:1 as well as international standard lipids. The supernatant was measured for metabolites by UPLC-MS/MS. The ultra performance liquid chromatography (UPLC, ExionLC AD), equipped with Thermo Accucore™ C30 column (i.d. 2.1 x 100 mm, 2.6 μm) and maintained at 45°C, was used for UPLC-MS/MS analysis (Wuhan Metware Biotechnology Co., Ltd., Wuhan, China).
