To identify effector genes of pancreatic beta cell dedifferentiation and dysfunction, we integrated analyses of histone methylation as a surrogate of gene activation status and RNA expression in beta cells sorted from mice with multiparity-induced diabetes. We performed H3K4me3 ChIPseq and H3K27me3 ChIPseq in FAC-sorted beta cells from female mice. There are a total of 6 conditions: 3-month-old nulliparous WT and RIP-Cre/FoxO1 floxed (IKO) mice, 12-month-old nulliparous WT and IKO mice, and 12-month-old multiparous WT (labeled as NGT) and IKO (labeled as DM) mice.
