The reproductive tracts of female wasps were excised under PDS. The calyces were then punctured with forceps causing the calyx fluid to diffuse in to the surrounding PBS, which was then collected and centrifuged at 3000 rpm for 3 min at 4 °C to precipitate eggs and cellular debris. The suspend calyx fluid was filtered through a 0.45 μm syringue filter to get filtered virus. The filtered virus was centrifuged at 4 °C for 10 min at 15,000 rpm. MbBV DNA was isolated from viral particles. The quality had been measured with NanoDrop to make sure 260/280>2.0, concentration > 15 μg/μl.
