Enhanced biological phosphorus removal (EBPRs) is the most reliable wastewater treatment method widely relying on microbial communities to remove phosphate from wastewater. Despite their stability and widespread usage, little is known about their microbial composition and activity. In the study, high-throughput pyrosequencing of bacterial community from two full-scale EBPR reactors of South Africa was investigated. For microbial analysis, unfiltered replicates were homogenously mixed and a 50 mL aliquot was taken, and centrifuged at 10 000 x g for 5 min at 4 ⁰C and the cell pellets harvested were re-suspended in 1x TE buffer (pH 8.0). The suspensions were well mixed and bacterial DNA was extracted with the ZR Fungal/Bacterial DNA KitTM (Zymo Research Corporation, USA) according to the procedures provided by the manufacturer. The integrity and purity of the metagenomic DNA was later assessed on the 0.8% agarose gel and Nanodrop spectrophotometer (Nanodrop 2000, Thermo Scientific, Japan). The PCR reaction was performed on all the extracted metagenomic DNA samples using the universal primer 27F and 518R.
