MeRIP was performed using a Magna Methylated RNA Immunoprecipitation Kit m6A Kit (Merck Millipore, USA). All the experimental procedures were carried out in accordance with the manufacturer's instructions. Overall design: Surgical samples were obtained from glioma patients and patients with traumatic head injury (normal tissues). To assess the difference in the m6A-modified transcriptome between glioma tissues and normal tissues, m6A-meRIP followed by high-throughput sequencing of 3 paired glioma and normal tissues was performed. MeRIP-seq and data analysis were performed by Shanghai Genechem Co., Ltd. We reveal abnormalities in the m6A epitranscriptome in glioma via MeRIP-seq, and our results emphasize that ALKBH5 mediates the upregulation of SH3RF3-AS1 for its stabilization and that SH3RF3-AS1 further exacerbates the malignant biological behavior of glioma cells through upregulation of SH3RF3.
