RNA-seq of trnascriptome from human embryonic stem cells treated with DMSO or 1 uM of RG7834

We aimed to investigate whether treatment with a small molecule drug RG7834 significantly affected the transcriptome of human embryonic stem cells carrying a pathogenic mutation in the gene DKC1. Overall design: RNA in triplicate from DMSO or RG7834 treated DKC1 hESCs was used for strand-specific library preparation using Nextflex Rapid Directional RNA-seq kit 2.0. Libraries were multiplexed and seqeunced on Illumina HiSeq using 2x150 bp reads. Demultiplexed reads were checked for quality, followed by adapter trimming, alignment to hg38 p13 version 99 from Ensembl and read counts were obtaiend using HTseq. Read counts were used for differential gene expression analysis using DESeq2. We did not find any significantly affected genes between the two conditions.
