Although Mycobacterium tuberculosis (Mtb) strains exhibit nucleotide level similarity of >99%, similarity is rarely replicated into the phenotype. Phenotypic heterogeneity in Mtb has been mostly associated to epigenetic inheritance. The most common epigenetic mechanism in Mtb is DNA methylation. At present the methylome of Mtb has not been completely characterized, neither has any resulting information been applied in inferring differences in clinical phenotypes observed in TB patients. Understanding the complete biology of Mtb will help in developing strategies for improving the Mtb treatment duration of the standard 6 months. This is the characterization of methylomes of 18 Mycobacterium tuberculosis (Mtb) isolates from patients in Blantyre, Malawi. This work presents the largest number of Mtb genomes to be completed in a single study using Single Molecule Real Time (SMRT) sequencing to date.
