Dynamics of calcineurin-regulated gene expression in the absence of Hog1 in S. cerevisiae

Gene expression was quantified in crz1 hog1 delete S. cerevisae cells over a time course following calcineurin activation by CaCl2. Cells were pretreated with buffer or the calcineurin inhibitor FK506 to allow the identification of calcineurin-regulated genes. We show that calcineurin-dependent downregulation of G1/S genes is partly dependent on the osmostress-activated kinase Hog1.
本研究对经氯化钙(CaCl₂)激活钙调神经磷酸酶(calcineurin)后的时间进程中,敲除CRZ1与HOG1的酿酒酵母(S. cerevisae)细胞内的基因表达水平进行了定量检测。实验设置缓冲液预处理与钙调神经磷酸酶抑制剂FK506预处理两组对照,以实现钙调神经磷酸酶调控基因的筛选鉴定。结果证实,钙调神经磷酸酶依赖的G1/S期基因下调过程,部分依赖于渗透压应激激活的激酶Hog1。
