Single cell rna seq raw data of Shank3B KO mice. Mus musculus

For the scRNA-seq experiments, brain samples from 2 control and 2 Shank3B deletion 8 week-old male mice (all 4 male mice) were prepared in the same single-cell isolation batch to minimize potential batch effects. The SVZ region tissues from control and Shank3B deletion mice were then dissected. The dissected tissues were then minced into 1 mm3 pieces using sterile razor blades and detached by incubation for 20 min in a solution containing 0.01% papain (25 U/mg, Worthington Biochemicals), and 0.01% DNase I (2788 U/mg, Worthington Biochemicals). The cell suspension was combined with an equal volume of 0.5% BSA + DPBS and filtered through a 70 um strainer, mixed with 4 ml of a debris removal solution (Miltenyi), and pelleted via centrifugation (3,000 g for 10 min). Myelin debris was then removed, and cells were separated and diffused in 1 ml of 0.5% BSA + DPBS. We pooled cells from wild type cells into a single 10X lane and the mutant cells into a single 10x lane.
