Matricaria recutita alpha-bisabolol synthase (MrBAS) promoter

alpha-bisabolol has high value because of its healthy function and medical value, and has a wide application prospect. A 1407 bp promoter upstream of the translation initiation site of the alpha-bisabolol synthase (MrBAS) gene (MrTPS1, Gene Bank No. KJ020282) was cloned from the genome of Matricaria recutita by TAIL-PCR chromosome walking technique. The alpha-bisabolol content and MrBAS expression level in different flower tissues showed that the expression of MrBAS was highly correlated with alpha-bisabolol content. 5 'deletion analysis showed that MrBAS promoter sequence could drive GUS gene expression in tobacco leaves, and its activity decreased with the decrease of promoter fragment. GUS staining and activity determination of transgenic Arabidopsis showed that MrBAS promoter could drive GUS gene expression in anthers, pollen tube and petal of Arabidopsis, and the expression was tissue specific. MrBAS has the potential to be developed as a specific tool to guide the specific expression of transgenic genes in flower organs in plants, and provide new research ideas for the biosynthesis of alpha-bisabolol and the development of new varieties with high alpha-bisabolol content.
