HS-AFM movies of RuvC–DNA interactions under +Mg²⁺ and −Mg²⁺ conditions

This dataset contains high-speed atomic force microscopy (HS-AFM) movies of Escherichia coli RuvC interacting with plasmid DNA under two experimental conditions, in the presence (+Mg²⁺) and absence (−Mg²⁺) of magnesium ions. The repository includes 35 individual cruciform-binding event movies extracted from full-length HS-AFM recordings (15 events under +Mg²⁺ conditions and 20 events under −Mg²⁺ conditions). In addition, full-length HS-AFM movies containing non-cruciform binding events used for the statistical analyses in Figures 3C and 5C are provided; these recordings include 127 non-cruciform binding events under +Mg²⁺ conditions and 126 under −Mg²⁺ conditions, with multiple binding events potentially occurring within a single movie. The repository further includes the complete raw dwell-time datasets (in seconds) used for the statistical analyses in Figures 3C and 5C, the raw cruciform structural classification counts (rabbit-ear, cross-shaped, and no-cruciform) for Figure 1D, and the raw particle height measurements for Figure S4.
