DMS-seq of maturational change in viral chromatin in human Adenoviruses

Adenovirus is a human pathogen that causes mild to severe ilness predominantely in children. Recently it has gained attention in being used as a vector for gene therapy and vaccination, yet much is yet unknown when it comes to its maturation during virion genesis. Here, Adenovirus particles were sequenced with DMS-seq to interrogate the organisational change of viral nucleoproteins inside the virion upon maturation. Samples consists of free DNA, Adenovirus C serotype 5 wildtype with a deletion in E3, and the immature Adenovirus 5 ts1 mutant with deletions in E1 and E3. As DMS is able to penetrate the viral capsid, it is the ideal agent to study the organisation within the virion and interrogate the difference between the wt and the ts1 mutant.
