PBMC were isolated from one individual. mRNA was extracted using standard Trizol method. First strand of cDNA were generated using primers specific for BCR’s C-segments. Universal adapter was introduced using reverse transcriptase template switching effect on 5' end of cDNA molecules. cDNA was amplified in two steps of PCR. To the final PCR product Illumina TruSeq adapters were ligated using convenient protocol.
