Overexpression of human progastrin (hGAS) in mice has been observed to increase colonic mucosa proliferation significantly, but the mechanism and receptors of progastrin have not been defined. We investigated this mechanism by observing the microarray gene expression in mGAS-/- mice, WT mice, and hGAS transgenic mice. Gene expression of colonic mucosa of WT C57BL/6 mice,Gas-/- C57BL/6 mice, and hGAS transgenic mice, all 6 weeks old were compared.
已有研究观察到,在小鼠体内过表达人胃泌素原(human progastrin, hGAS)可显著促进结肠黏膜增殖,但胃泌素原的作用机制及其对应的受体尚未明确。本研究通过检测mGAS敲除(mGAS-/-)小鼠、野生型(wild type, WT)小鼠及hGAS转基因小鼠的基因芯片表达谱,探究上述作用机制。本研究对6周龄的野生型C57BL/6小鼠、Gas-/- C57BL/6小鼠以及hGAS转基因小鼠的结肠黏膜基因表达水平进行了对比分析。
