Employing multi-omics analyses to understand changes during kidney development in perinatal Interleukin-6 animal model

Chronic kidney disease (CKD) is a leading global health concern. Maternal obesity during pregnancy leads to increased levels of the pro-inflammatory cytokine IL-6, impacting fetal development. Administering IL-6 or saline to pregnant mice during mid-gestation, we analyzed kidney tissue from newborns using mRNA-seq, miRNA-seq, and whole-genome bisulfite-seq. Employing a multi-omics approach, we identified 19 key genes regulated by epigenetics and miRNAs. Constructing a regulatory network, we found genes affecting pathways such as Mannose type O-glycan biosynthesis, Cell cycle, and Apoptosis. Notably, Atp7b and Man2a1 genes were influenced by DNA methylation and miRNA targeting, suggesting a role in fetal programming linked to CKD development from gestational inflammation.
