The microbial composition in the cecum of pig’s influences host health, immunity, nutrient digestion, and feeding requirements significantly. Advancement in metagenome sequencing technologies like 16S rRNA provided an extra edge to determine cecum microbial population. In this study, we performed a comparative analysis of crossbred Korean native pig cecum microbiota at two different stages (Stage L=10 week, and Stage LD=26 week) with the help of 16S rRNA sequencing technology. Our results revealed remarkable differences of microbial composition, α and β diversity, and differential abundance between two stages. Phylum composition analysis with respect to SILVA132 database showed presence of Firmicutes is 45.97 % and 40.99 % in stage L and LD respectively. Similarly presence of Bacteroidetes was 43.5 % in stage L and 52.20 % in LD. The genera Prevotella, Anaerovibrio, Succinivibrio, Megasphaera were differentially enriched in stage L, whereas Clostridium, Terrisporobacter, Rikenellaceae were enriched in stage LD ( ). Functional annotation of microbiome by level three KEGG pathway analysis revealed that glycine, serine, threonine, valine, leucine, isoleucine arginine, proline , and tryptophan metabolism are differentially enriched in stage L, whereas alanine, aspartate, glutamate, cysteine, methionine metabolism, phenylalanine, tyrosine and tryptophan biosynthesis metabolism are differentially enriched in stage LD. Through machine learning approaches like LEfSe (Linear discriminant analysis Effect Size), Random forest, Pearson’s correlation, we found pathways like amino acid metabolism, transport systems, and genetic regulations metabolism are commonly enriched in both stages. Our findings suggest that the bacterial compositions in cecum content of pigs are highly involved in their nutrient digestion process.Keywords: cecum microbiota, 16S rRNA sequencing, SILVA database.
