Mapping replication timing in the RPE1 and HCT116 cell lines revealed differences in replication dynamics and replication domain structures in the two cell lines Overall design: Asynchronously growing cell populations were pulsed with the thymidine analogue 5-Ethynyl-2'-deoxyuridine (EdU) and sorted inot an early, mid and late - S phase fraction. Nascent DNA was isolated from each fraction and sequenced. RPE 1 experiments were performed in duplicate.
