We were interested in studying the role of DOT1L-mediated H3K79 methylation on N-Myc target gene transcription in MYCN-induced neuroblastoma. We aimed to determine whether knocking down DOT1L in MYCN-amplified neuroblastoma would affect N-Myc target gene expression. We generated neuroblastoma BE(2)-C cells stably expressing control (SCRAMBLE) or DOT1L shRNA and treated the cells with control vehicle (DMSO) or doxycycline to induce DOT1L expression knockdown for 96 hours.
