We sequenced 28 Mlh1-/- and 28 Mlh1+/+ mouse T cells together with bulk DNA samples from thymic T cells to study the mutational landscape shaped by replication errors and DNA mismatch repair (MMR). Mlh1 is an integral part of the MMR machinery, and Mlh1-/- mice are defective of MMR.
