Data have been deposited on the development of mixed-mode dispersive solid phase extraction for therapeutic, modified oligonucleotides. Five silica-based adsorbents functionalized with aminopropyl, aliphatic, aromatic, and dicarboxylic groups were prepared and thoroughly characterized. A novel mixed-mode dispersive solid-phase extraction method was established using a central composite experimental design, allowing systematic investigation of the parameters influencing antisense oligonucleotide desorption. The performance of the method was evaluated using oligonucleotides that differed in length and chemical modifications. The developed approach was successfully applied to the extraction of antisense oligonucleotides from cerebrospinal fluid, providing consistent recoveries ranging from 40% to 93%. Matrix effects were minimal. The best recoveries were achieved for oligonucleotides modified at two structural sites.
