Small non-coding RNAs (sncRNAs), including microRNAs and piRNAs, play crucial roles in regulating gene expression in cancer, influencing key processes such as cell proliferation, apoptosis, and metastasis. This also includes piR-1254, a 32 nucleotides long non-coding RNA. Here it is shown that piR-1254 has a broad influence on the expression of more than 2000 different genes. The expression of half of the genes is reduced and that of the other half is increased. To investigate the influence of piR-1254 on differential expression in the Neuroblastoma cell line Kelly, the cells were reverse transient transfected with piR-1254 or piR-Ctrl as control. 48 h after transfection, the RNA was harvested and a total RNA-seq was performed.
