We characterized the diet of the Punta Banda Estuary harbor seal colony (Mexico:Baja California) through metabarcoding. Briefly, we used a two-step PCR method, included the V9 region in eukaryotes and 16S mitochondrial region of Chordata and cephalopods species. A blocking oligonucleotide matching harbor seal sequences was used in the first PCR to prevent seal DNA amplification.
