Nuclear extracts were isolated from leaves of ZmHsfA5-myc OE lines at the three-leaf stage. samples were collected at 45 °C for 0 h and 45 °C for 3 h, respectively. Each biological replicate comprised 1.0 g leaf tissue (n = 2 replicates per group). Protein-DNA complexes were captured using the a CUT&Tag® assay kit (TD901-01, Vazyme). The experimental group employed an anti-myc antibody (ab32072, Abcam, UK), while whereas the control group used an anti-IgG antibody. Sequencing libraries were constructed following manufacturer's instructions and sequenced with 150-bp paired-end reads on an Illumina NovaSeq 6000 platform (GENEWIZ, Suzhou, Jiangsu, China).
