We profiled transcriptomes in human lung cancer cell line H23 when the expression of HOTAIR was knockdown by the siRNA specific to an HOTAIR isoform HOTAIR-N (NR_047517). Overall design: H23 cells were cultured in RPMI1640. HOTAIR-N-specific and control siRNAs were transfected into H23 cells using RNAiMAX. Total cell RNA was extracted at 48 hrs after transfection. Three independent transfections were carried out for each siRNA. The extracted RNA was processed for RNA-SEQ on Illumina HIGHSEQ 2500.
