Platinum drugs have broad spectrum and high anti-cancer activity with nucleus DNA as primary target. During platinum chemotherapeutics, the steady-state levels of reactive oxygen species (ROS) are generally elevated and induce oxidative stress response, to regulate cell survival and drug resistance. Currently, a novel nucleus-to-mitochondria ROS pathway has been identified. However, the pathways by which platinum drugs generate ROS, as well as the biological response, still lack systematic studies. Using carboplatin as an example, we systematically analysed the dynamic response to ROS in cancer cells. Overall design: To thoroughly understand the effects of platinum chemotherapeutics on the expression of the global transcriptome, we performed the mRNA sequencing (mRNA-Seq) on the MCF-7 cells treated with or without 0.5 mM carboplatin. NC group: Normal MCF-7 cells, C group: carboplatin treated MCF-7 cells.
