This repository contains single-molecule FRET data and Matlab analysis code related to the manuscript "MreC-MreD structure reveals a multifaceted interface that controls MreC conformation" (Figures 2 and 3). 1) Matlab code (FilerTrajectoriesForEbFRET.m) that filters and formats single-molecule trajectories for further ebFRET software processing (van den Meent et al. Biophys J 2014, https://ebfret.github.io/), using outputs from a previously published raw movie processing pipeline (Graham et al. Mol Cell 2016).  2) Outputs from ebFRET processing are provided in the “ebFRET outputs” folder, organized by figure and condition. The three outputs are: 1) the ebFRET session file (.mat file), 2) a list of all single-molecule trajectories (.dat file), scored by the smFRET state assigned to each timepoint (typically 1-2),  and 3) a summary file (.csv file) with statistics of the smFRET distributions and their dynamics. The ebFRET session file can be loaded directly into ebFRET gui to visualize the trajectories and the resulting FRET efficiency distributions. The latter two files can be visualized and analyzed using either custom code, or the example Matlab code provided here (PlotEbFRETOutput.m). 3) FRET efficiency data that were used to construct smFRET histograms are provided in the “smFRET histograms.xlsx” file, organized by figure and condition.
