Chinese inbred line 654, developed by the Chinese National Sorghum Improvement Center at the Liaoning Academy of Agricultural Sciences (LAAS), is a high-yielding grain sorghum with favorable agronomic traits, including photoperiod insensitivity, early maturity, short internodes and a thin stem, and dwarfism. It has been used as a parent line for numerous commercial hybrids in China, and also as a parent for recombinant inbred lines (RILs), which have been instrumental in cloning several agronomically important traits such as grain size and color. Here, we present a telomere-to-telomere (T2T) genome assembly and well-annotated genes of line 654 using raw reads including ultra-long ONT, PacBio HiFi, Hi-C, and RNA-Seq.
