A complete hydatidiform mole (CHM) is a common disease known to develop post-molar gestational trophoblast neoplasia (GTN). However, the details of the molecular biological mechanisms underlying the progression to post-molar GTN from hydatidiform moles are still largely unknown. In this study, we performed RNA sequencing (RNA-seq) using archival fresh frozen tissues of patients with CHM and compared the gene expression profiles of patients who achieved spontaneous remission and those who developed post-molar GTN after the evacuation. Overall design: We prospectively collected CHM tissues from patients who underwent the evacuation at the Nagoya University Hospital (Nagoya, Japan). In this study, we included seven patients with CHM who eventually developed low-risk GTN between 2015 to 2021. Moreover, we included six patients with CHM who achieved spontaneous remission as controls. The total RNA of their archival fresh frozen tissues was extracted using the miRNeasy®? Mini Kit (QIAGEN, Hilden, Germany), and the library construction and pair-end sequencing were conducted by Rarevariant, Inc. (Tokyo, Japan).
