Skeletal muscle gene expression analysis of Shank3delta11(-/-) and wildtype mice at postnatal day 0-1

We analyzed transcriptional differences by RNA-sequencing of muscle tissue of neonatal Shank3delta11(-/-) mice and compared those to Shank3(+/+) controls. We found significant differences in gene expression of ion channels crucial for muscle contraction and for molecules involved in calcium ion regulation. In addition, calcium storage- (i.e. Calsequestrin (CSQ)), calcium secretion- and calcium- related signaling- proteins were found to be affected. We used RNA-seq data from five Shank3delta11(-/-) and four wildtype mice skeletal muscle to perform gene expression profiling.

我们通过RNA测序(RNA-seq)分析新生Shank3delta11(-/-)小鼠的肌肉组织转录差异,并与Shank3(+/+)野生型对照小鼠进行比对。结果显示,两类小鼠中,调控肌肉收缩的关键离子通道以及参与钙离子调控的分子,其基因表达存在显著差异。此外,钙存储相关蛋白(即肌集钙蛋白(Calsequestrin, CSQ))、钙分泌相关蛋白及钙信号通路相关蛋白均出现表达异常。本研究使用5只Shank3delta11(-/-)纯合子小鼠与4只野生型小鼠的骨骼肌RNA测序数据,完成了基因表达谱分析。
