In this study, RNA-seq was used to detect mRNA changes in DI-AKI cell models induced by three drugs (Iohexol, cisplatin and vancomycin), and was analyzed and validated to screen key hub genes and specific biomarkers for DI-AKI. Overall design: This study was divided into four groups of samples with three replicates in each group. The first group was the control group, and water was the control lysozyme. The second group was iohexol induced-AKI. Iohexol (75 mg I/mL) was used to intervene HK-2 cells for 48h. The third group was the cisplatin induced-AKI group. The HK-2 cells were treated with cisplatin (5 µM) for 48h. The fourth group was vancomycin induced-AKI.Vancomycin (4 mM) was used to intervene HK-2 cells for 48h.
