Here we utilized large-scale systematic probing and screening of ~2000 sequence and structural variants based on two long, perfect RNA hairpins to explore the structure and sequence context determining editability. Overall design: 2 replicates of B2 or mNG first arm amplicon libraries from basal HEK293T, IFN-a stimulated HEK293T, siAdar treated HEK293T, MCF7, A549 and NIH3T3 cells. 2 replicates of B2 second arm amplicon libraries from basal HEK293T, IFN-a stimulated HEK293T and MCF7 cells. 1 sample each of poly-A selected library of HEK293T cells transfected with B2 or mNG positive control reporter constructs.
