To test whether TSLP triggers the homeostatic maturation program of human splenic cDC2 (DC2hm program) in vitro, we administrated TSLP to in-culture human splenic cDC2 and performed RNA-seq. The results showed that TSLP induced DC2hm-like profile including the upregualtion of genes related to DC maturation, migration, immunoregulation, Treg induction and did not induce drastic ISG upregulation compared to either freshly isolated or cDC2s cultured in medium. Overall design: Purified cDC2s were treated with TSLP (50 ng/ml) or complete medium (Med) for 12h . UID RNA-seq which incorporating unique identifiers to improve sensitivity and remove PCR-ampliaction bias was then performed on TSLP-cDC2s, Med-cDC2s and purified cDC2s.
