Effect of loss of Jmjd1c on normal hematopoiesis

Analysis of gene expression profile of LSK (Lin-Sca-1+ c-Kit+) isolated from Jmjd1c f/f Vav1Cre or Vav1Cre controls. Loss of Jmjd1c minimally affected HSC in homeostatsis while impairs HSC function in response to stress such as transplantation and 5-Fu treatment. These results provide insight into the role of Jmjd1c in normal hematopoiesis. LSK cells were isolated from bone marrow of seven weeks old Jmjd1c f/f Vav1Cre or Vav1Cre controls mice by flow activated cell sorting and subjected to RNA-seq.
本研究对从Jmjd1c f/f Vav1Cre小鼠及Vav1Cre对照小鼠中分离得到的LSK细胞(Lin⁻Sca-1⁺c-Kit⁺)开展基因表达谱分析。Jmjd1c缺失对稳态条件下的造血干细胞(Hematopoietic Stem Cell, HSC)功能仅产生轻微影响,却会损害其在移植、5-氟尿嘧啶(5-Fu)处理等应激状态下的功能。上述研究结果为阐明Jmjd1c在正常造血过程中的作用提供了重要见解。本研究通过流式细胞分选术,从7周龄Jmjd1c f/f Vav1Cre小鼠及Vav1Cre对照小鼠的骨髓中分离LSK细胞,并对其进行RNA-seq分析。
