The UPS is an important regulator of cell signaling and proteostasis. Ubiquitin protein ligase E3 component n-recognin 5 (UBR5), a HECT-domain E3 ubiquitin ligase, is emerging as a key regulator of the UPS in cancer and development. Dysregulations, including amplification and mutation, were commonly found in various cancers, however, the underlying mechanism is poorly understood. Recently, using organoid based single cell CRISPR screening, we identified Ubr5 as the regulator of hepatocyte differentiation and maturation. However, the mechanism of how Ubr5 control liver cell fate determination has not been fully investigated yet. Here, we performed snRNA-seq and bulk RNA-seq of control and Ubr5-KO livers to systematically explore it functions in liver.
