Droplet based single cell RNA sequencing profiles of a murine model of type-1 diabetes

Diabetic kidney disease remains the leading cause of end-stage renal disease, affecting ~30% of patients with long-standing type 1 and type 2 diabetes, and is characterized by proteinuria and gradual loss of kidney function. There is intense interest in understanding the responses of the many cell types present in the kidney to the diabetic milleu. In this study, we present single cell transcriptomes of renal cortex samples of wild-type and diabetic (C57BL/6-Ins2Akita/J) mice using Drop-Seq. Renal cortex samples from wild type and diabetic Akita mice (n=2 per group) underwent dissociation and Drop-seq.

糖尿病肾病(Diabetic kidney disease)仍是终末期肾病(end-stage renal disease)的首要病因,累及约30%的长期1型糖尿病(type 1 diabetes)与2型糖尿病(type 2 diabetes)患者,其特征为蛋白尿(proteinuria)与肾功能进行性减退。当前学界对阐明肾脏内多种细胞类型在糖尿病微环境中的应答反应有着浓厚的研究兴趣。本研究采用Drop-Seq技术,获取了野生型(wild-type)与糖尿病型(C57BL/6-Ins2Akita/J)小鼠肾皮质(renal cortex)样本的单细胞转录组(single cell transcriptomes)数据。本研究中,野生型与糖尿病Akita小鼠的肾皮质样本(每组各2只)均经过细胞解离处理后,开展了Drop-seq测序。
