To measure mRNA levels of inflammatory cytokines tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β), and interleukin-6 (IL-6), total RNA was extracted from substantia nigra tissues using the RNeasy Mini Kit (QIAGEN). cDNA synthesis was performed with Superscript III Reverse Transcriptase (Fermentas), and quantitative real-time PCR was used to detect cytokine mRNA levels. β-actin served as an internal control for normalization. Gene expression was calculated with the 2−ΔΔCT method. Primer details are in <b>Table 1</b>.
