O-GlcNAcylation is the modification of serine and threonine residues with beta-N-acetylglucosamine (O-GlcNAc) on intracellular proteins. To investigate the role of protein O-GlcNAcylation on intestinal homeostasis, we generated intestinal epithelial cell (IEC)-specific O-GlcNAc transferase (OGT) knockout in mice. The KO mice developed spontanous intestinal inflammation. To determine the underlying molecular mechanisms, we performed RNA sequencing of ileum and colon epithelial cells of wildtype and IEC-OGT KO mice. mRNA profiling of intestinal epithelial cells (IECs) from the ileum and colon of 10-week-old wildtype and IEC-OGT KO male mice.

O-连接β-N-乙酰葡糖胺修饰(O-GlcNAcylation)是一类发生于细胞内蛋白质丝氨酸与苏氨酸残基、以β-N-乙酰葡糖胺(O-GlcNAc)为修饰基团的共价翻译后修饰。为探究蛋白质O-连接β-N-乙酰葡糖胺修饰在肠道稳态中的调控作用,本研究构建了肠上皮细胞(IEC)特异性O-连接β-N-乙酰葡糖胺转移酶(OGT)敲除小鼠模型。该敲除小鼠可自发出现肠道炎症表型。为解析其潜在分子机制,本研究对野生型与IEC-OGT敲除小鼠的回肠、结肠上皮细胞开展了RNA测序(RNA sequencing)。本数据集包含10周龄雄性野生型与IEC-OGT敲除小鼠的回肠、结肠肠上皮细胞的mRNA表达谱分析数据。
