Genome-wide identification of target genes for the histone demethylase Phf8 in the oligodendroglial cell line CG4 [Phf8KO]

To identify target genes of the histone demethylase Phf8 in oligodendroglial cells in a genome-wide and unbiased approach, RNA-sequencing studies were performed on wildtype CG4 cells and CRISPR/Cas9-edited Phf8 knockout cell lines. We compared three wildtype and one CRISPR/Cas9-treated clone without genome editing with four CRISPR/Cas9 Phf8-knockout clones. Among others we detected substantial changes in the expression of genes associated with cell cycle and replication of DNA.为在全基因组范围内以无偏倚的实验策略鉴定少突胶质细胞中组蛋白去甲基化酶Phf8的靶基因，我们对野生型CG4细胞与经CRISPR/Cas9编辑的Phf8敲除细胞系开展了RNA测序（RNA-sequencing）研究。我们比较了3株野生型CG4细胞、1株经CRISPR/Cas9处理但未进行基因组编辑的克隆，以及4株CRISPR/Cas9介导的Phf8敲除克隆。本研究中，除观测到其他多项表达变化外，我们还检测到与细胞周期及DNA复制相关的基因表达发生了显著改变。
