RNA-Seq of purple potato tubers in different development stage

A tetraploid potato genotype with purple color of tuber skin and flesh Huasong 66 kindly provided by HuaSong Seed Industry (Beijing) co LTD (Inner Mongolia Autonomous Region China) was used in the present study. Sowing was carried out on the farm of Agricultural College of Inner Mongolia Agricultural University on Early May 2018 In the potato developmental phases, timely weeding fertilization and watering Potato tubers of five different developmental phases (sa and sb tuber formation period sc and sd tuber expansion period se starch accumulation period 2/3 of the stems and leaves above ground withered ) were collected between July 29th and September 7th 2018 at 10 days intervals Last sampling (sf maturing stage all the stems and leaves are withered) were collected at September 26th 2018 Total RNAs were extracted from purple potato tuber using the TRK1002 total RNA purification kit according to the manufacturers instructions Equal volumes of RNA extracts from Sa to Sf were pooled for use as S1 or S3 for sRNA library construction and sequencing Three libraries were denoted S1 (sa and sb) S2 (sc sd and se) S3(sf)
