Metastasis is one of the defining features of pancreatic ductal adenocarcinoma (PDAC) that contributes to poor prognosis. In this study, the palmitoyl transferase ZDHHC20 was identified in an in vivo shRNA screen as critical for metastatic outgrowth, with no effect on proliferation and migration in vitro, or primary PDAC growth in mice. In this experiment, we sought to determine the changes in the transcriptome of Zdhhc20 KO cells that were previously generated using CRISPR/Cas9 and com,pare them with control cells. This experiment comprises three groups, with 3 biologial replicates each. The Zdhhc20 KO cells (clone E4) were generated using CRISPR/Cas9, whereas Control cells (Clone C5, WTCtrl) have also undergone nucleofection with the same gRNAs, but without successful editing and were used as a single-cell cloned control that expresses Zdhhc20. The parental cells (PDA50 Met, WT) were used as an additional control group.
