A single RNA sequencing approach using 10x Genomics scRNAseq was employed to stuudy induction of anegy in hunam T lymphocytes. Anergy is a condition of immune T cells becoming sustainably inactive. Theoretically, it occurs when a T cell recognizes its antigen but cannot be active. Induction of anergy by single amino acid mutated antigen has been shown in mouse models decades ago. The present study applied this method to human cells in vitro, and demonstrated induction of annergy in human T lymphocytes. Overall design: HLA-A24-positive healthy donor derived PBMC was cultured with (A24-3) or without (A24-1) single amino acid mutated CMV-antigen (QYDPVSALF) for 24hrs, followed by culture in the presence of the original CMV-antigen (QYDPVAALF) for two weeks.
