Photoconverted microglia single cell RNA sequencing

This project investigates the initial immune responses of microglia to disseminated tumor cells (DTCs) during the early stages of brain metastasis. Using a transgenic Cx3cr1-PSmOrange2 mouse model, we combined intravital two-photon imaging with targeted photoconversion (Opto-omics) to isolate tumor-adjacent microglia. Photoconverted microglia and control distant microglia were collected by FACS, and single-cell transcriptomes were obtained using SMART-Seq v4 and sequenced on Illumina NovaSeq. The dataset provides raw FASTQ files of single-cell RNA sequencing and reveals transcriptional heterogeneity of microglia in the tumor microenvironment. These data will contribute to understanding microglial plasticity and identifying novel therapeutic targets for early intervention in brain metastasis.
