Effect of MS-275 and CXD101 on gene expression of M-MDSCs and PMN-MDSCs [RNA-Seq]

To elucidate the molecular mechanisms of HDAC1-3 inhibitors in inhibiting M-MDSCs, RNA-seq analysis of M-MDSCs and PMN-MDSCs isolated from Hepa 1-6 tumor-bearing mice treated with MS-275 or CXD101 were performed. Overall design: M-MDSCs (CD11b+Ly-6ChiLy-6G-) and PMN-MDSCs (CD11b+Ly-6CintLy-6G+) were sorted by JSAN (KS-Techno, Japan) from tumor isolated. Total RNA was extracted from M-MDSCs or PMN-MDSCs using miRNeasy Micro Kit (Qiagen). RNA libraries were prepared using TruSeq stranded mRNA sample prep kit (Illumina, USA) and sequenced on Illumina NovaSeq 6000 platform platform (101-base single-end mode).
