The impact of canagliflozin on gene expression changes was investigated in human renal proximal tubular cells (HK2) after pre-treatment with high glucose. Human renal proximal tubular cells (HK2) were cultured in Keratinocyte-Serum Free Medium containing 10% fetal bovine serum (FBS), 5 ng/ml recombinant epidermal growth factor (rEGF), 0.05 mg/ml bovine pituitary extract (BPE), 100 U/ml penicillin and 100 µg/ml streptomycin. All cells were grown at 37 °C in a humidified atmosphere with 5% CO2. After growth to confluency, cells were serum deprived for 24 hours. Thereafter, cells were either treated with 0.5 µM canagliflozin [n=4] or left untreated [n=4] and subsequently stimulated with high glucose (30 mM).
