The aim of this study was to find correlates of high antibody titers in COVID-convalescent plasma (CCP) donors and to study the time course of transfused and endogenous antibody titers in COVID-19 patients.
Here, we studied plasma samples from 93 CCP donors collected up to 174 days after symptoms ended. These CCP donors had mild disease. In addition we collected plasma from 16 severe COVID-19 patients before and after CCP transfusion. We measured anti-SARS-CoV-2 Spike RBD, S1, and N-protein antibodies in all samples using ELISA. In addition, capacity of antibodies to block ACE2-RBD binding was assessed. Viral N-protein and viral RNA were measured in plasma of the 16 COVID-19 patients as an estimate for their viral load.
