We report the application of RNA sequencing technology for high-throughput profiling of transcriptome in mammalian cells. In this study, to search the potential candidate genes that may responsible for the GSK343-mediated promotion of MDSC generation, hematopoietic stem cells(HPCs) were isolated from bone marrow of C57BL/6 mice, then, treated with or without GSK343 upon MDSC-induction. 24 hours later, cells were harvested for RNA sequence analysis. We find that GSK343 promotes HPCs to differentiate into MDSCs by activating Jak-stat3 and TNF signaling pathway.
