Spike-in CUT&Tag was performed against the cofactor Chip in control and after optogenetic nuclear depletion in 2-4h, 6-8h, and 10-12h Drosophila embryos. It was also performed against CBP, H3K27ac, H3K18ac, H4K8ac, BRD4, and Zelda in control and depleted 2-4h embryos, and against Chip and Zelda in control and zelda knockout 2-4h embryos. Experiments were performed in duplicates and nuclei from Drosophila virilis were used as a spike-in.
